Work­ing field:

Exper­i­mentelle Radi­olo­gie, Col­lab­or­at­ive Research Cen­ter (CRC) SFB 1340 "In vivo Visu­al­iz­a­tion of Extra­cel­lu­lar Mat­rix Patho­logy" (Mat­rix in Vis­ion), Pro­ject B03 "Tar­get­ing Extra­cel­lu­lar Mat­rix Changes Induced by an Altered Homeo­stasis of Phos­phate Meta­bol­ism". Within this pro­ject a doc­toral pos­i­tion is avail­able.

The sum­mar­ized hypo­thesis for this pro­ject is that uremic car­di­omy­opathy (UC) is accom­pan­ied by a patho­lo­gic increase of the glyc­osa­minoglycan (GAG) com­pon­ent of the myocar­dial extra­cel­lu­lar mat­rix (ECM) and that these changes can be detec­ted and char­ac­ter­ized by in vivo Mag­netic Res­on­ance Ima­ging (MRI) using appro­pri­ate cationic ima­ging probes as well as by elas­t­ography-based in vivo mag­netic res­on­ance ima­ging (MRI).

  • Exper­i­mental work with cell cul­tures (e.g. THP cells, fibro­cytes) under vary­ing phos­phate con­cen­tra­tions of the medium, ana­lysis of the pro­duced ECM-com­pon­ents, incub­a­tion with vari­ous cationic ima­ging probes and ana­lysis of the bind­ing of these probes to ECM com­pon­ents.
  • In vivo research using a model for UC (adenin fed small animal). par­ti­cip­a­tion in and sup­port of in vivo ima­ging using small animal MRI, pre­par­a­tion of the tis­sue samples (heart and other organs) for ex vivo MRI and 1H relax­o­met­rie, and for his­to­chemic and immun­o­his­to­chemic micro­scopy. Fur­theron pro­cessing of tis­sue samples for bio­chem­ical ana­lysis, espe­cially for char­ac­ter­iz­a­tion of ECM com­pon­ents.
  • Assist­ance in the invest­ig­a­tion of bind­ing of cationic iron oxide nan­o­particles and Gad­olinium com­pounds to GAGs using spec­tro­scopic and relax­o­met­ric meth­ods.
  • Assist­ance in the invest­ig­a­tion of the vis­coelastic prop­er­ties of tis­sues using multiscale elas­t­ography.
A spe­cial focus will be on on the elu­cid­a­tion of the struc­tures of GAGs and how these struc­tures cor­rel­ate with the bind­ing of ima­ging probes and the dis­ease pro­cess.
