Work­ing field:

An adverse pren­atal envir­on­ment can have long-term effects on the health of the child, which can mani­fest much later in life and lead, for example, to the devel­op­ment of res­pir­at­ory dis­eases. The molecu­lar mech­an­isms trans­mit­ting early expos­ure effects to the phen­o­typic scale are incom­pletely under­stood, and appro­pri­ate in vitro sys­tems are miss­ing. We are par­tic­u­larly inter­ested in per­sist­ent epi­gen­etic modi­fic­a­tions and their func­tional trans­la­tion in response to such early expos­ures that might help to under­stand the devel­op­ment of res­pir­at­ory com­plic­a­tions. This Ph.D. pro­ject will com­ple­ment our epi­demi­olo­gical stud­ies by estab­lish­ing and apply­ing in vitro mod­els for dif­fer­ent early adverse expos­ure scen­arios.

The job holder will be part of a research group focus­ing on epi­gen­etic modi­fic­a­tions and health effects. In his/her pro­ject, the job holder will estab­lish in vitro mod­els to invest­ig­ate the con­sequences of chem­ical/par­tic­u­late mat­ter expos­ure. He/she will estab­lish lung organoid cul­tures to study the prim­ing effects of an adverse envir­on­ment dur­ing devel­op­ment. Bey­ond organoid cell cul­ture, dif­fer­ent molecu­lar bio­logy and ima­ging tech­niques will be applied, includ­ing but not lim­ited to e.g., pyrosequen­cing and qPCR. Single-cell RNA sequen­cing stud­ies will com­ple­ment epi­gen­etic stud­ies.
